Edward M. Bednarczyk, PharmD

Research interests are focused to the application of molecular imaging techniques such as positron emission tomography (PET) to assessing drug effects, including the study of migraine headache where we have looked at treatment as well as induction of headache. We have also had recent work looking at opiate receptors in the human brain using 11C-carfentanil, a radiopharmaceutical that binds to the mu opiate receptor.

 

The characteristics of positron emitting radio nuclides often make them ideal tracers for measuring drug effects. This includes the labeling drugs with PET radio nuclides such as C-11, or F-18. These nuclides allow for creation of labeled drugs nearly identical to their unlabelled counterparts. We have also used ³workhorse¹ radiopharmaceuticals such as FDG in a variety of settings ranging from measuring the uptake of glucose into white blood cells, to oral administration of this radiopharmaceutical, to application in the assessment of response to drugs in dementia. Other research areas include migraine headache, and neuroreceptor binding studies.

Linda Catanzaro, PharmD

Director of the Pharmacotherapy Information Center.

Primary research interests are in the fields of drug information and pharmacy informatics. Currently, Dr. Catanzaro is continuing to work in collaboration with other UB SoPPS faculty on projects with the NY State Department of Health (DOH). The projects involve post-marketing drug utilization literature evaluation as well as retrospective drug utilization review (DUR) research for the Office of Medicaid Clinical Drug Review Program. Additional projects for this year will involve the development and tracking of outcomes for the DOH Prescriber Education Program (PEP) as well as the development of a web-based Rapid Response Center for providing drug information in support of the PEP within the Pharmacotherapy Information Center. 

 

Students interested in this type of research will have the opportunity to develop their literature retrieval and evaluation skills and professional writing skills. The DUR research component will provide the opportunity to learn how to analyze insurance claims databases to identify prescribing patterns and opportunities for optimizing appropriate medication use. Participation in the project may also lead to the opportunity for the student to become a published author.

Alice Ceacareanu, PharmD/PhD

Research focuses on elucidating the mechanisms of cancer invasion induced or exacerbated by nitric oxide and hallmarks of diabetes type 2. Dr. Ceacareanu’s laboratory also aims at developing predictive screening tools to forecast cancer recurrence and guide antineoplastic therapy to improve treatment outcomes.

Dr. Demler is the principle investigator on the following research protocols:

  1. Aripiprazole augmention, is an effective approach to psychiatric pharmacotherapy
  2. Safety profile associated with the concomitant use of lorazepam with olanzapine, ziprasidone or haloperidol
  3. Safety profile and efficacy associated with the concomitant use of
  4. clozapine and benzodiazepines.
  5. Clinical outcomes if concomitant use of oseltamivir and clozapine; assessing changes in neutrophil and white blood cell counts
  6. Comorbid illness in a psychiatric state hospital

 

Future research interests include evaluating efficacy and safety of current psychiatric pharmacotherapeutic interventions, with potential to improve long term success, cost effectiveness and patient adherence.

Research interests in cardiology, pharmacoeconomics, outcomes research, and health services research.  Specific areas of research include cost-effectiveness of pharmacogenetics and clinical pharmacy services as well as patient preference and decision-making.

  1. Research and application of the laws, rules and regulations, both federal and state affecting pharmacy practice issues in NYS. These include current legislative issues affecting the expansion of pharmacists' responsibility in patient care along with the application of business models for improving the corresponding revenue streams that any legal changes may create.  
  2. Studying areas to improve the recognition, and reporting of medication errors and systems for improvement of practice sites for enhancement of patient outcomes.  A New York State Department of Health pilot study involving medication errors and prescriptions is currently underway. 
  3. Management of adult Asthma in the ambulatory/community settings.  I am a board certified educator in asthma and am working with the Western New York Asthma Coalition.
  4. Business plan modeling for area pharmacy related businesses to improve efficiency and profitability.

Research focuses on three areas: 1) characterization of drug transporter proteins and evaluation of protein function on the intracellular disposition of HIV-1 PIs in peripheral blood mononuclear cells between the maternal and fetal compartments; 2) evaluation of maternal, pregnancy and newborn outcomes following antepartum chemoprophylaxis in HIV-infected women; 2) pharmacokinetics/pharmacodynamics of ART in pregnancy; and 3) HIV-1 protease inhibitors induced placental cell injury; and 4)

 

Strong efforts are made to characterize a variety of drug efflux transporter proteins and its function in the intracellular disposition of HIV-1 PIs in PBMCs between the mother and baby at the time of delivery.

 

Data on the safety of ART use and obstetrical management of ART-associated side effects during pregnancy remain limited.  A database to monitor maternal, pregnancy, newborn outcomes study following antepartum ART chemoprophylaxis to reduce perinatal HIV-1 transmission has been established for patients referred to the HIV High Risk Pregnancy Program at the Ohio State University.  Students will have opportunities to establish and maintain an outcomes database for HIV-infected pregnant women referred to the Erie County Medical Center (ECMC).  Efforts will be made to merge the outcomes database between ECMC and OSU.  The inter-university collaboration between OSU and the University at Buffalo (UB) will increase the outcomes data to better assess the efficacy and safety of ART use during pregnancy.

 

We recently reported a significant increase in the placental weight, placenta: newborn weight ratio, surface area, and thickness in dams despite a lack of difference in maternal plasma glucose.  A similar trend was observed among HIV (+) women on a PI-containing antepartum regimen.  These clinical findings support those observed in the experimental rat model and suggest a PI effect on the placenta that is glucose-independent.  We are directing our research efforts on exploring PI-induced placental endothelial cell injury as a possible mechanism.

Associate Director of the UB NYS Center of Excellence for Bioinformatics and Life Sciences.

Dr. Morse leads the UB AIDS Clinical Trials Group Pharmacology Specialty Laboratory and the UB HIV/AIDS Clinical Pharmacology Quality Assurance and Quality Control Program that are supported by NIH grants from the National Institute of Allergy and Infectious Diseases and the National Institute on Drug Abuse. Dr. Morse directs the HIV Treatment Education Program at the Erie County Medical Center in Buffalo and the International Center for HIV/AIDS Pharmacotherapy Research and Training Program (ICHAPRT). ICHAPRT provided research and education for international students and faculty with an interest in HIV/AIDS treatment. Information on these programs is available at www.hiv.buffalo.edu.

Dr. Morse, along with other SoPPS faculty, is developing the Translational Pharmacology Research Core (TPRC) program in the New York State Center of Excellence in Bioinformatics and Life Sciences. The TPRC has four components including: 1) a Translational Pharmacology Core Facility for Bioanalysis and Drug Development, 2) a Bioinformatics and Medication Management Research Network, 3) a Translational Pharmacogenomics program and 4) a Translational Pharmacotherapy Research Center. The TPRC has been designated by the Patient Safety Center of the NYS Department of Health as the Coordinating Center for a statewide ePrescribing initiative. These research components have common areas of interest with the other research cores and SoPPS faculty research initiatives. The TPRC is actively participating in the UB 2020 planning process with other faculty and clinician scientists located on the Buffalo Niagara Medical Campus to develop interdisciplinary programs within the UB 2020 Health and Wellness Across the Lifespan Strategic Strength.

Research interests lie in the area of cardiovascular disease with a specific focus on antiplatelet pharmacology and platelet function monitoring.  There is increasing interest in the use of platelet function tests to monitor the effects of antiplatelet drugs, with the goal of guiding therapy to the optimal dose for the prevention and treatment of thrombosis while minimizing hemorrhagic complications.  In addition, the cardiovascular risk of several non-cardiac drugs has recently come into question.  Dr. Norgard’s research incorporates the coated-platelet assay, an innovative approach for analyzing platelet function and the thrombotic potential of a drug.

 

Research goal is to provide critical insight into the coated-platelet role in thrombotic occlusions that may ultimately be utilized to design effective strategies for prevention and treatment of atherothrombotic diseases and to establish the coated-platelet assay as a marker of thrombotic capability that can be used to monitor medication efficacy and safety.

This clinical research program has been an ongoing collaborative program with the Division of Nephrology at Erie County Medical Center for over 15 years focusing on the pharmacokinetics, pharmacodynamics and pharmacogenomics of immunosuppressive agents during renal transplantation:

 

Current and upcoming clinical research endeavors focus upon pharmacokinetics of immunosuppressive drugs in relationship to pharmacodynamics of immunologic markers and targeted pharmacogenomic endpoints in relation to race and gender of the renal transplant recipient.  Research projects may provide the student with exposure to a variety of biomedical technologies including LCMSMS, flow cytometry and Q-PCR. These projects provide Pharm.D. students with an opportunity to explore clinical and translational research projects though a clinical research team (i.e. Pharm.D.s, physicians, nurse clinicians, immunologists, geneticists, biostatisticians, etc.) by participation in clinical pharmacology sub-studies in renal transplant patients with the endpoint to provide safe and efficacious immuosuppression (e.g. cyclosporine, prednisone, mycophenolic acid) among different patient groups.

 

In addition, this funded clinical research program has focused on the pharmacokinetics and dynamics of glucocorticoids with specific emphasis on the impact of the factors of gender, race, acute rejection, time post-transplant, immunologic response and chronic adverse effects in the renal transplant population. This research program has also evaluated the pharmacodynamics of anti-lymphocyte induction agents as well as glucocorticoid pharmacokinetics during various “steroid withdrawal” protocols during a multi-center clinical trial.

Brian Tsuji, PharmD

Research interests involve investigating of mechanisms of resistance in multi drug resistant bacteria and pharmacokinetic/pharmacodynamic modeling of antimicrobials to suppress the emergence of resistance. Dr. Tsuji’s interest in antimicrobial resistance mechanisms involves the utilization of novel in vitro pharmacodynamic models and investigation of bacterial genetic factors which contribute to drug resistance.

Research interests focus on the care of Hospice and Palliative Care patients.  This includes investigating the impact of Clinical Pharmacy Services (CPS) on the care of patients as well on the pharmacoeconomic impact on the providers and payors. Specifically, clinical investigations include approaches to opioid use and pain management strategies and use of novel products and delivery systems in patients experiencing nausea and vomiting.

Joseph P. Balthasar, PhD

Director, Center for Protein Therapeutics with research focusing on the utilization of pharmacokinetic and pharmacodynamic analyses and mathematical modeling to guide the discovery and development of new immunotherapies for cancer and autoimmunity.

Research Interests and Projects:

1.      Development of new therapies for immune thrombocytopenia (ITP).  This laboratory has developed new animal models ITP, and these models now allow systematic, mechanistic investigation of new therapies for this common autoimmune disease. Current research is investigating the therapeutic utility of: (a) “Opsisomes” (antibody-coated liposomes) that are designed to block pathways of platelet destruction inITP, (b) an extracorporeal bioreactor that is designed to remove pathogenic autoantibodies, and (c) “Macro-block” a new therapy that is designed to utilize endogenous antibodies and a triggering agent to form immune complexes that block the phagocytic activity of macrophages.

2.      Development of FcRn inhibitors.  This project focuses on the design and evaluation of inhibitors of FcRn, a transport protein that protects IgG from intracellular catabolism. The inhibitors may have broad utility in the treatment of antibody mediated auto- and allo- immune conditions.

3.      Targeting tissue plasminogen activator (tPA). This project will be performed in collaboration with Dr. Victor Yang at the University of Michigan. Research conducted at UB will investigate the utility of novel delivery strategies for selective alteration in the pharmacokinetics and pharmacodynamics of tPA.

4.      Targeting chemotherapy with novel derivatives of the TAT peptide. This project represents another collaborative effort between UB and Dr. Yang’s group at the University of Michigan. Research conducted at UB will investigate the utility of novel delivery strategies that attempt to enhance the pharmacokinetic, pharmacodynamic, and therapeutic selectivity of anti-cancer drugs.

5.      Inverse targeting for ovarian cancer. This project employs anti-drug antibodies to alter the distribution and elimination kinetics of chemotherapeutics as a means to provide targeted drug therapy of peritoneal metastases of ovarian cancer. Our approach combines intraperitoneal administration of antineoplastic agents with systemic administration of anti-drug antibodies. The presence of anti-drug antibodies in the systemic circulation leads to rapid binding of drug diffusing out of the peritoneum and entering the blood, thereby reducing the extravascular distribution of the chemotherapeutic. It is anticipated that resultant alterations in the pharmacokinetics of the chemotherapeutic will lead to a reduction in the fraction of drug delivered to tissues associated with toxicity (e.g., bone marrow), without affecting peritoneal drug exposure or peritoneal cytotoxic activity.  Thus, this drug targeting approach may beneficially alter the efficacy v. toxicity profile of antineoplastics utilized in the treatment of ovarian cancers.

 

Sathyamangalam Balu-Iyer, PhD

Research interest is in the area of pharmaceutical biotechnology, in particular lipid nano/microparticulate mediated delivery of biotechnology derived products such as therapeutic proteins and siRNA.

Current research projects:

(1)   Development of lipidic nano particle containing therapeutic proteins and is supported by NHLBI/NIH. The overall goal of the project is to develop less immunogenic and long acting therapeutic proteins to improve safety and efficacy of protein based products. We adopt multidisciplinary approach involving Biophysics, immunology and pharmacokinetics/Pharmacodynamics.

(2)   Re-activating Memory T Cells in the Microenvironment of Human Tumors and development of in situ vaccination. This project is supported by NCI/NIH ((Dr. Bankert, PI, Balu-Iyer Co-PI). Our aim is to develop lipidic Microparticulates containing therapeutics such as IL-12 that can reverse TCR arrest using Biophysical, immunological and Toxico kinetics approaches.

(3)   Design lipid based particle to overcome three distinct barriers in si RNA delivery that include (a) Improve in vivo stability; (b) Target organs and cell type and (c) Facilitate intracellular delivery.

(4)   Formulation and delivery of Monoclonal antibody based products: We adopt rational strategies based on protein folding approaches to develop stable and efficacious formulations for monoclonal antibody based products.

 

Javier Blanco, PhD

Research focus is the improvement of cancer chemotherapy by understanding the factors that contribute to interindividual variability in drug response. Dr. Blanco’s goal is to perform translational research using a combination of approaches based on a) the analysis of biological samples from selected populations, and b) the use of informative laboratory models.

 

A project in Dr. Blanco’s laboratory is related to the understanding of the factors that govern interindividual variability in the metabolism and disposition of the anthracyclines doxorubicin and daunorubicin. It is possible that specific sequence variations in genes associated with the metabolism and disposition of anthracyclines may impact the risk of cardiotoxicity in some individuals. Our efforts are focused on the discovery and characterization of novel single-nucleotide polymorphisms in a family of genes involved in the metabolism of anthracyclines in liver, heart and different types of tumors (e.g. breast and lung cancer). We are investigating the extent of DNA sequence variation in these selected gene candidates among normal individuals from different ethnic groups. We will study genotype-phenotype correlations by using paired tissue-DNA-RNA samples available from my collaborators from the Pharmacogenetics of Anticancer Agents Research group (PAAR, http://www.pharmacogenetics.org/), and from the Roswell Park Cancer Institute (RPCI, http://www.roswellpark.org/document_9.html). We will expand our studies to characterize the functional effect of the genetic variants by using different in-vitro and in-vivo models (e.g. cultures of hepatocytes, cardiomyocytes, transgenic mice).

 

Our findings are being translated into informative case-control epidemiological studies. I am a co-investigator in the project entitled “Cardiac events among survivors of childhood and adolescent cancer – Role of gene-environment interactions” (PI: Smita Bhatia, City of Hope National Medical Center, CA). The project is sponsored by the Childhood Cancer Survivor Study Group (The Children’s Oncology Group http://www.childrensoncologygroup.org/), and supported by a grant from the Lance Armstrong Foundation. We are analyzing the distribution of the candidate polymorphisms in identically treated patients who have vs. have not developed congestive heart failure (CHF) after anthracycline treatment for childhood cancer. In the future, individualizing the dosing of anthracyclines based on genetic characteristics might minimize the occurrence of adverse effects.

 

Kathleen MK Boje, PhD

Research interests include:

  1. Neuroinflammatory Brain Diseases.  Understanding the role of neuroinflammatory processes in the progression of chronic neurodegenerative diseases.  Since inflammatory processes are a common feature of many neurological diseases (viz., Alzheimer's disease, multiple sclerosis, HIV-1 dementia, cerebral ischemia, brain tumors and meningitis), an enhanced knowledge of inflammatory mediators and their detrimental effects on the central nervous system provides opportunities for the design of new pharmaceutical approaches in the management of neurological diseases.
  2. Drug transport across the blood-brain barrier.  Focuses on the mechanisms of drug transport across the blood-brain and blood-cerebrospinal fluid barriers.  Nature designed these barriers to restrict and regulate the entry of blood borne substances into brain tissue.  Essential nutrients are transported efficiently by carrier proteins expressed by the blood-brain and blood-cerebrospinal fluid barriers.  However, these barriers hinder the entry of many drugs, and typically it is only highly lipophilic drugs that gain access to brain tissue via passive diffusion across the barriers.  Consequently, many lead drug candidates are disqualified from further development because of poor permeability across the blood-brain barrier.  There is much scientific interest in understanding brain transport processes with the goal of identifying methods, which enhance drug delivery across brain barriers.
  3. N-Methyl-D-Aspartate Receptors.  Explore the pharmacologic modulation of the functional activity and expression of brain N-methyl-D-aspartate receptors (NMDA).  The NMDA receptor system is an important glutamate receptor subtype within the central nervous system. Appropriate receptor activation is critical for neuronal development and differentiation, synaptic plasticity, learning and memory processes, whereas receptor over activation initiates or contributes to neurotoxicity and neurodegenerative diseases.  Hence, an understanding of the neuropharmacology of NMDA receptors may lead to the development of new drugs for disease management.

 

Daniel A. Brazeau, PhD

Director, Pharmaceutical Genetics Laboratory (PGL).  The roll of the PGL is to provide extensive training and research support for a wide array of molecular genetic techniques to assist researchers in addressing a broad range of questions including gene expression, bioinformatics and genotyping.  Students are encouraged to visit the PGL if they are interested in research opportunities in pharmacogenomics or pharmacogenetics.  The laboratory has the necessary equipment and expertise to conduct a broad spectrum of techniques in molecular genetic biology including gene expression studies (mRNA Differential Display, northern analysis, microarrays, quantitative real-time PCR) and genotyping (RFLP’s, AFLP’s, ISSR’s, and SNP’s).  In addition the PGL provides extensive education and training to not only university students and faculty, but also to outside researchers and professionals through a series of workshops and short courses in DNA microarray technology, statistical packages/programs for genetic analysis, microsatellite development and analysis, and molecular genetic methodologies for the pharmaceutical sciences.   

 

To aid students and faculty in their research the PGL has all the necessary equipment for the isolation, characterization and cloning of DNA/cDNA(mRNA) including three PCR thermocyclers and a Nanodrop ND-1000 Spectrophotometer for measuring DNA/RNA concentrations in small volumes (1 µl).  For gene expression studies the laboratory has a separate room (165 sq ft), certified for isotope use with hybridization chambers, incubators, water baths, refrigerator, and centrifuge.  The laboratory has a new Kodak large-format image analysis system (Kodak 2000MM Multimodal Imaging System).  For scanning of microarray slides the lab has a GenePix 4000A (Axon Instruments).   The lab also has an Mx3005P multiplex quantitative PCR system (Stratagene) for gene expression studies and high-throughput genotyping.  The laboratory also has a Zeiss Axiovert 220 inverted microscope with DIC and fluorescent capabilities

 

Gayle A. Brazeau, PhD

Research focuses on investigating the interactions of drugs, molecules and diseases with skeletal muscle from a perspective of characterizing and understanding the mechanisms responsible for tissue damage and/or pain. There are three types of projects that are being investigated in the laboratory. In one area, we have been investigating the mechanisms of tissue damage and/or pain associated with components in parenteral formulations. This work is conducted using a variety of experimental techniques ranging from cell culture to isolated tissues to whole animal studies. In related studies, we have been involved with the development of in situ formulations that cause minimal tissue damage and can provide long-term drug delivery. A second area is focusing on elucidating the mechanisms responsible for pain upon injection. The overall goal is to investigate the link between tissue damage with pain upon injection and to develop excipients or drugs that minimize or eliminate pain or tissue damage upon injection.

 

We have now investigating the role of myostatin in muscle wasting conditions. A second major focus is to investigate the underlying mechanisms responsible for alcoholic myopathy. Alcoholic myopathy and/or cardiomyopathy is known to occur in 1/3 to 2/3 of those who ingest alcohol on a chronic basis. An understanding of these mechanisms can be used as a basis to develop therapeutic approaches to minimize these toxic effects in cardiac or skeletal muscle. We are also investigating the mechanisms associated with increased susceptibility to muscle and cardiac damage and decreased muscle performance associated with reduced estrogen levels seen in young women, post-menopausal women and in aging. These studies have shown beneficial effects of estrogen, but the mechanisms, either a membrane or receptor mediated, are not clearly elucidated. Alcoholic myopathy and the potential beneficial effects of estrogen on muscle are investigated using our available animal models. In more recent studies, we have been investigating the role myostatin may play as a mediator for muscle growth and/or wasting.

Dr. Fung is specifically characterizing genetic causes of the development of tolerance to organic nitrates and seeks methods of maximizing the formulation stability and delivery of organic nitrates for treatment of various cardiovascular disorders.

Research interests include nuclear receptor biology and drug metabolism, with focus on the on the structural biology and molecular mechanism of actions of nuclear receptors and coregulators, molecular pharmacology of selective nuclear receptor modulators, and nuclear receptor-mediated xenobiotic response in drug metabolism.

 

William J. Jusko, PhD

Research interests are in theoretical, basic, and clinical aspects of the pharmacokinetics and pharmacodynamics of various immunosuppressive agents including corticosteroids, as well as drugs used to treat diabetes, inflammation, and cancer. With Drs. Richard Almon and Debra DuBois, Dr. Jusko delves into the pharmacogenomics of diverse effects of corticosteroids and has evolved advanced mathematical models of receptor/gene-mediated responses.  They have characterized the effects of corticosteroids on hepatic and muscle enzymes and tissue responses and have evolved advanced PK/PD models for cascade-type processes. Dr. Jusko has developed mechanism-based pharmacokinetic, pharmacodynamic, and disease progression models and computational methods describing the action of various drugs and utilizes mathematical models of drug action to determine optimal dosage regimens diverse drugs.

 

Wojciech Krzyzanski, PhD

Research interest focuses on the application of mathematical modeling as a tool to explore functioning of complex biological systems regulated by processes that can be controlled pharmacologically. Dr. Kryzanski’s primary interest is in the hematopoietic system, attempting to reach a balance between his theoretical work in the broadly understood field of pharmacometrics and experimental studies.

  1. Hematopoietic system: Therapeutic applications of hematopoietic growth factors (erythropoietin, thrombopoietin, and G-CSF) and their recombinant human analogs. Toxic effects of anticancer agents on hematopoietic cells in blood and bone marrow. Dynamics of hematopoietic cell populations. Receptor mediated disposition of hematopoietic growth factors.
  2.  Pharmacometrics: Development through mathematical analysis and computer simulations of complex indirect response models, in particular, those which describe cell kinetics driven by lifespan distributions. Application of these models in population PK/PD modeling of agents affecting various cell populations.

Research interest areas:

  1. Integrative and Systems Pharmacology of Anti-Platelet and Anti-Cancer Drugs
  2. Development and experimental validation of quantitative structure-pharmacokinetic/pharmacodynamic (PK/PD) relationships (QSPR) to optimize drug design, development, and therapeutic application of anti-cancer compounds.
  3. Integration of QSPR and mechanistic PD models developed with several drugs to predict in vivo PK/PD profiles of chemically related compounds.
  4. Development of models of pharmacological target-mediated drug disposition and dynamics, and the experimental validation of the primary determinants of PK/PD profiles of drugs that exhibit such phenomena.
  5. Development of non-deterministic models such as neural networks, alone or in combination with traditional adaptive feedback control methods, for individual optimization of complex pharmacotherapeutic regimens.
  6. Incorporation of biomedical signal processing tools into PK/PD models for characterizing the pharmacology of drugs that alter spontaneous variability of signals of the cardiovascular and autonomic nervous systems.

 

Marilyn E. Morris, PhD

Research interests are focused in the areas of drug transport and pharmacokinetics/pharmacodynamics (PK/PD) of drugs.  Current research projects:

  1. Gamma-hydroxybutyric acid (GHB) Toxicokinetics and Toxicodynamics.  GHB is a drug of abuse, known as a “club drug”.  We are characterizing the mechanisms underlying the toxicokinetics and toxicodynamics of GHB, and evaluating methods to treat overdoses of GHB.  For these studies, we are characterizing transport of GHB by Monocarboxylic Acid Transporters (MCTs), transport proteins that determine the absorption, renal clearance, and distribution of GHB throughout the body, including its distribution to the brain, the site of action.  Our goal is to devise treatment strategies to treat overdoses of GHB, in order to save lives. 
  2. Multidrug Resistance in Breast Cancer.  Drug resistance is the main cause for therapeutic failure and death in breast cancer.  My laboratory is interested in the discovery of new classes of compounds useful in reversing multidrug resistance (MDR) to cancer chemotherapeutic agents.  One of the main causes of MDR in cancers is due to the overexpression of the efflux transport proteins, P-glycoprotein (P-gp) and Breast Cancer Resistance Protein (BCRP).  Our current studies are evaluating various dietary components, organic isothiocyanates and flavonoids, for their effects on BCRP-mediated transport and on the efficacy and toxicity of chemotherapeutic agents. 
  3. Cancer Preventive Agents. We are investigating the effects of dietary flavonoids and organic isothiocyantes on cancer initiation and progression, and the use of these natural products in cancer prevention.  We are interested in the changes in gene expression produced by these dietary compounds and how these relate to cancer prevention.  
  4. Biliary Excretion and Enterohepatic Cycling of Drugs.  Our research focuses on the importance of the ABC transport proteins in the biliary excretion of drugs, the in silico and in vitro methods for the prediction of biliary clearance in humans, and modeling enterohepatic cycling of drugs.  

 

Jun Qu, PhD

Research focus areas: Proteomics and Pharmaceutical Analysis. Major research programs in the proteomic field involve i) high-resolution and large-scale expression profiling of pathological proteomes (e.g. for cardiovascular diseases, colon cancer and infectious diseases) for the discovery of disease/therapeutic biomarkers by gel-free LC/MS methods; ii) Sensitive identification, localization and quantification of post-translational modifications in complex proteomes, with the emphases on arginine methylation and phosphorylation. Novel anti-PTM-peptides capture procedure and alternating collision induced dissociation (CID)/electron transferring dissociation (ETD) are employed to obtain abundant PTM information; iii) targeted quantification of regulatory, marker proteins for clinical study. Dr. Qu’s lab possesses many state-of-the-art LC/MS instruments, including a high resolution/accuracy LTQ/Orbitrap XL with ETD, a highly sensitive TSQ Quantum Ultra EMR triple-quadrupole instrument, two ultra-high pressure nano-LC systems, and several HPLC instruments for pre-fraction and ion chromatography.   A number of key analytical advances have been developed by his lab that greatly enhanced the proteomic coverage, sensitivity and throughput for proteomic research. As for the Pharmaceutical Analysis of small-molecule drug/markers, Dr. Qu’s lab is focusing on the ultra-sensitive quantifications of drug, metabolites and endogenous markers (e.g. corticosteroids, di-hydroxyl-vitamin D metabolites, androgens, etc.) using a novel combination of selective enrichment and micro- or nano- LC/MS.

 

Murali Ramanathan, PhD

Research interests are in the area of treatment of multiple sclerosis (MS), an inflammatory-demyelinating disease of the central nervous system that affects over 1 million patients worldwide.  MS is a complex, variable disease that causes physical and cognitive disability and nearly 50% if patients diagnosed with MS are unable to walk after 15 years.  The etiology and pathogenesis of MS remains poorly understood.  The focus of the research is to identify the molecular mechanisms by which the autoimmunity of MS is translated into neurological damage in the CNS.

 

Current research focuses on work in the following patient-centered MS projects:

  1. Molecular mechanisms of disease and treatment effects in MS and other demylelinating diseases.
  2. Therapy optimization
  3. Gene expression profiling and proteomics
  4. Mathematical models for pharmacogenomic and signal transduction processes and immunomodulatory protein networks.

 

Jerome J. Schentag, PharmD

Research interest focuses on the effects of acute and chronic diseases on the PK/PD of antibiotics and cardiovascular agents.  These studies are primarily conducted in phase II-IV clinical trials of antibiotics and other therapeutic categories.  Dr Schentag’s research organization is dedicated to the conduct of Adaptive Clinical Trials incorporating PK/PD methodology into drug development. 

 

Robert M. Straubinger, PhD

Dr. Straubinger’s research program involves the application of drug carriers for the treatment of infectious diseases and cancer. Current efforts are directed toward improving the therapy of brain tumors (as well as others) by targeting tumor blood vessels. We employ a variety of experimental approaches, including high-resolution magnetic resonance imaging, liquid chromatography/mass spectroscopy, confocal fluorescence microscopy and image analysis, pharmacokinetic/dynamic analysis, and molecular techniques such as quantitative RT-PCR. With these techniques, we examine the effects of treatment upon tumor vascular permeability and drug deposition, the localization of the carrier-delivered drug within the tumor, and the molecular mechanisms involved when tumor blood vessels are attacked during therapy.

 

Additional interests of the lab that are being pursued actively include understanding the mechanisms of anti-cancer drug action and how they may be modified by changing exposure profile (time vs. concentration). Both genomic and proteomic analysis approaches are utilized in this work. 

 

Aiming Yu, PhD

Research interests lie in the field of drug metabolism and disposition. Dr. Yu’s laboratory is interested in studying the function and regulation of human cytochrome P450 (e.g., CYP2D6 and CYP3A4) drug-metabolizing enzymes and ATP-binding cassette (ABC) membrane transporters (e.g., BCRP/ABCG2 and MRP1/ABCC1), and the consequent effects on pharmacokinetics and drug responses. The ultimate goal is to identify the molecular mechanisms of interindividual variability in response to xenobiotic drugs that supports the discovery and development of new drugs, and contributes to a more rational drug therapy.

 

  1. Role of small noncoding RNAs in the regulation of drug metabolism and disposition in cancer therapy. Understanding the function of small RNAs in regulation of ABC transporter expression shall provide new insight into multidrug resistance, which may offer clue for the development of novel strategy to combating drug-resistant cancers.
  2. Pharmacogenetics in the metabolism of indolealkylamine agents and drug-drug interactions. Delineation of the genetic and environmental factors that affect indolealkylamine metabolism, pharmacokinetics and drug effects will advance the understanding of risk factors in use of psychoactive indolealkylamine drugs.
